About five miles south of Biggersville, Rienzi was named for Nicola Gabrina Rienzi, a Roman Tribune, and has been at its present location since 1860. Previously the old town had been one mile west of its current location, scattered about the old Nesly Williams Plantation, which was settled in 1830.
When old Tishomingo County was divided into Alcorn, Prentiss, and Tishomingo Counties, Rienzi fell just inside Alcorn County.
